Concerning this PBPK model for nicotine:
http://tobaccodocuments.org/rjr/521808687-8698.htmlWe can see in the last page (p186) that the renal clearance (determined very classically with Ae/AUC) is implemented directly in the kidney, on the venous blood concentration (Cvk).
My question is : is it correct from a theoretical point of view?
I mean I thought that this clearance (estimated from the systemic plasma concentration) should be applied on the systemic plasma concentration. Am I wrong? If we apply it in the kidneys concentration, should we adapt it or not (considering that the renal blood flow is less than the total blood flow etc.)?
Currently, I implement it in the kidneys, but on the arterial kidney concentration (Ca), which is the systemic concentration. Am I wrong?
